Would that be a problem? Or perfectly alright?
I'm planning on finally putting a rig together for the first time, and trying to cut financial corners as I'm downright poor. lol The external HD that I'm talking about is a 160GB 7200 rpm Toshiba 3.5". Thanks in advance for any and all input!

Should not present a problem. You would treat it as a brand new drive and reformat. If this is a pre-assembled external enclosure I don' know if removing the drive will be an issue.

If you want to keep any data on it, then don't reformat it. If it's a second drive, just make sure you cable it correctly. Same if it's going to be the primary HD. You can install Windows on it without reformatting.
