What motherboard do your have? Have you checked the Windows Sound Properties? Are your speakers known-working?

Which port are you plugging the speakers into?

Are you using powered speakers?

Is the on-board audio enabled in the BIOS?

USB uses codecs and bypasses the sound card. Your sound card is not working, if it's enabled in the bios you have a bad driver, you are not using the correct jacks, or a setting is wrong in its control panel.
