A Screenshot of what ?

Windows ? -> PrtScr key (or ALT + PrtScr for active window)
DirectX game ? -> Via extra software. . .or sometimes the games themselves have a screenshot function
Video ? -> Playback Software

It should work. Do you get anything when you paste, or nothing at all ?

Pressing Print Screen copies the screen shot to your "clipboard". They you paste it into Word, Paint, Photoshop, etc... and save it.

Well, paste it into paint or whatever image application you use.

I'm sure there is. You don't get any notification, but still, the desktop does get captured.
