In theory a thumb drive's lifespan depends on the number of times it's written to (though I notice that Crucial is now giving one of theirs a lifetime guarantee).
In practice how many times can it be dropped or stepped on or put through the washing machine before it just quits?
Destruction by owner if far more likely.
So yeah if you feel you have to you can defrag the drive.

Defragging a thumb drive is not recommended.

I have had multiple flash drives and I know got a pocket external hard drive. I have found that my drives have fragmented a little bit, but not enough to slow down. If you do defragment, then I recommend a program called Diskeeper. I use it on all my workstations and servers. It fast, powerful, and simple to use.
